Cell Prep

Sasha Bakhru

Remove plated cells from substrate by trypsin treatment; once suspended, neutralize trypsin. Centrifuge cells, and once pelleted, decant supernatant fluid.

Resuspend in alginate rich medium and load into cartridge.

Next prime the system and position the cartridge above a receiving bath containing calcium rich media.

Trigger at low frequency to expel the cell suspension into the bath below.

The cells will then be contained within a calcium-alginate template capsule.

Next the cell/capsule systems are to be diluted 2:1 with a 300 mOsm sucrose solution.

Finally, a pH 7.2 buffered chitosan solution is to be introduced for calcium displacement.

After some time, the mixture is to be centrifuged, and the supernatant decanted and disposed of.

The remaining matter is to be resuspended in buffered minimal media and stored in a sterile environment for use.
